How bacteria resistance to drugs?
Tetracycline is mainly used for TB treatment. So as we know TB is highly capable of resistance, they are able to uptake gene codes from resistant bacterias and multiply. But U'm not very sure about the mechanism of how the bacteria prevents it from harm.
There are many mechanisms , you should ask how bacteria resist a certain family of drug , because some bacteria developed different ways to resist the action of that drug

I can comment on Penicillin, 1st of all Penicillin is highly effective for gram positive bacteria and less effective for gram negative bacteria. This is due to the cell wall layer of the bacteria. So basically Penicillin acts on the transpeptidase enzyme (which is the key enzyme for forming the bacterial cell wall). Some bacteria are able to bypass this process by having enzymes like Penicillinase(which destroys the Penicillin itself), some have altered PBP(Penicillin binding protein) which has lower affinity for Penicillin binding.
Some bacteria also produce beta lactamase which destroys beta lactam (found in Penicillin which has a structural analog to D-alanine and inhibits the formation of cell wall)
