ABCDE is the acronym used in assessing skin lesions that are suspect of being cancerous. A is for Asymetry, B is for Border (irregular), C is for Color (inconsistencies), D is for Diameter, & E is for Elevation (raised in any dimension above the base skin level)
Biopsy, but I think it has to be excision biopsy, not shave or punch if you are thinking Melanoma. Dermatology is not my speciality
