Gram Staining

Gram-positive bacteria have a thick peptidoglycan layer in their cell wall and thus retain the color of primary stain i.e crystal violet and appear purple blue. While in contrast, Gram negative bacteria have a high content of lipopolysaccharide in their cell wall which is easily decolonized by acetone alcohol and thus cells take up the color of secondary stain or counterstain and appear pink in color.
