Hi Im wondering if anyone can explain exactly how organophosphates inhibit acetylcholinesterase (AChE)? If you know of any journal articles or reputable websites with this information, I'd love to have a read. Thanks heaps :)
The binding site in the acetylcholinesterase has a higher binding affinity for the Sulfur on the organophosphate nerve agents than the Oxygen of the acetylcholine. By binding to the Acetylcholinesterase the nerve agent inhibits the binding of Acetylcholine preventing the muscle contraction signals to be stopped Immediate treatment with a Nerve Agent Antidote Kit (NAAK) made up of Atropine 2-Pralidoxime Chloride (provides better binding for the organophosphates than the AChe), Diazepam (anticonvulsant) if they're seizing, along with rescue breathing (chest compressions/chest arm lifts, etc.) can keep a warfighter alive. The U.S. Military did tests on Nerve Agents in the 1950s. There's a video where they perform this treatment on a goat that gets VX Nerve Agent applied directly to the eye
